Lahir dengan John Christopher Depp di Owensboro, Kentucky, dari pasangan John Christopher Depp, Sr. dan Betty Sue Wells, pada 9 Juni 1963, Johnny Depp dibesarkan di Florida. Dulu ia pernah kecanduan narkotik, Dia drop out dari sekolah pada usia 15 tahun dengan harapan untuk menjadi seorang musisi rock. Namun hidupnya berubah ketika Depp masuk ke akting setelah kunjungan ke Los Angeles, California, dengan mantan istrinya, Lori Anne Allison (Lori A. Depp), yang memperkenalkannya dengan aktor Nicolas Cage.
Dalam hal asmara, Johnny pernah menjalin asmara dengan aktris Hollywood yang merupakan lawan mainnya dalam EDWARD'S SCISSORHAND. Setelah hubungan mereka retak Johnny mengencani supermodel Inggris Kate Moss namun hubungan ini pun tidak bertahan. Hubungan asmaranya yang terakhir yaitu dengan penyanyi Vanessa Paradis, dan mereka telah dikarunia 2 anak tanpa pernikahan. Sebelum tinggal bersama dengan Vanessa, ia pernah menikah dengan Lori Anne Allison yang berprofesi sebagai make up artis. Rumah tangga pasangan ini tak bertahan lama. Keduanya bercerai setelah tiga tahun menikah.
Dia membuat debut film dalam A Nightmare on Elm Street (1984). Pada tahun 1987 adalah awal karir bintangnya ketika ia menggantikan Jeff Yagher dalam peran polisi yang menyamar Tommy Hanson dalam serial TV populer "21 Jump Street" (1987).
Pada tahun 1990, setelah banyak peran dalam film-film yang berorientasi remaja, yang pertama dari sekelompok besar kolaborasi dengan sutradara Tim Burton terjadi ketika Depp memainkan peran judul dalam Edward Scissorhands (1990).
